Click Chemistry Immobilization of Antibodies on Gold Nanoparticle

Click chemistry is a chemical tool that allow the instantaneous and quantitative ligation between two molecules. In a paper recently published on Langmuir, researchers from CNR-ISTM, Nanobiolab (University of Milan Bicocca) and Labion (FDG), this powerful chemical reaction has been applied to allow the stable conjugation of antibodies on the surface of gold nanoparticles.

Gold nanoparticles have been previously coated with a newly developed copolymer (DMA-PMA-MAPS) containing some alkyne functionality. This polymer, not only stabilizes the nanoparticles avoiding their unspecific interactions and aggregation, but is also able to quickly react with azido modified anti-mouse IgG antibodies.

The combination of gold nanoparticles with antibodies is extremely important in diagnostic. These combination is in fact at the basis of lateral flow test and is currently used in a number of biosensing techniques. More stable and active IgG-Gold nanoparticles could thus allow to improve the sensitivity of a number of diagnostic tests.
